چکیده

Bone marrow mesenchymal stem cells (MSCs) can differentiate into smooth muscle cells (SMCs) and have tremendous potential for cell therapy and tissue engineering. In this study, to understand the effects of TGF-β3 on rat bone marrow-derived MSCs and the underlying molecular mechanism of this differentiation process, we investigated that the changes of myocardin-related transcription factors (MRTFs) at the transcriptional level after rat MSCs were treated with TGF-β3. The results showed that TGF-β3 increased the expression of contractile genes, such as SM22, smooth muscle-myosin heavy chain (SMMHC), SM-α-actin in MSCs. When TGF-β3 induced MSCs differentiation into SMCs, myocardin and MRTF-A were activated. The data indicated that TGF-β3 induced rat bone marrow-derived MSCs differentiation into SMCs by activating mypcardin and MRTF-A.

Osteogenic Differentiation of Rat Mesenchymal Stem Cells from Adipose Tissue in Comparison with Bone Marrow Mesenchymal Stem Cells: Melatonin As a Differentiation Factor

Background: Adipose-derived stem cells (ADSC) could be an appealing alternative to bone marrow stem cells (BMSC) for engineering cell-based osteoinductive grafts. Meanwhile, prior studies have demonstrated that melatonin can stimulate osteogenic differentiation. Therefore, we assayed and compared the melatonin effect on osteogenic differentiation of BMSC with that of ADSC. Methods: Mesenchymal...

Effect of Lithium Chloride on Proliferation and Bone Differentiation of Rat Marrow-Derived Mesenchymal Stem Cells in Culture

Objective(s) It is believed that the mesenchymal stem cell (MSC) differentiation and proliferation are the results of activation of wnt signaling pathway. On the other hand, lithium chloride is reported to be able to activate this pathway. The objective of this study was to investigate the effect of lithium on in vitro proliferation and bone differentiation of marrow-derived MSC. Materials and ...

Multilineage Differentiation Activity by the Human Umbilical Vein-Derived Mesenchymal Stem Cells

Background: Mesenchymal stem cells (MSC) are a very promising transplantable stem cell source for a variety of cell replacement therapies. As the main source of MSC is bone marrow (BM), most of studies have been done on BM-derived MSC (BM-MSC). Umbilical cord (UC)-derived MSC (UC-MSC) which are recently introduced, is one of the good alternative source for these cells. The objective of this stu...

Sertoli cell condition medium can induce germ like cells from bone marrow derived mesenchymal stem cells

Objective(s): Although many researchers have confirmed induction of germ cells from bone marrow mesenchymal stem cells (BMMSCs), there are no reports that confirm spontaneous differentiation of germ cells from BMMSCs. In this study, we have evaluated the effect of adult Sertoli cell condition medium (SCCM) as a mutative factor in the induction of germ cells from BMMSCs. Materials and Methods: ...

Capillary Network Formation by Endothelial Cells Differentiated from Human Bone Marrow Mesenchymal Stem Cells

Human bone marrow derived mesenchymal stem cells (HBMSCs) have the potential to differentiate into cells such as adipocyte, osteocyte, hepatocyte and endothelial cells. In this study, the differentiation of hBMSCs into endothelial like-cells was induced in presence of vascular endothelial growth factor (VEGF) and insulin-like growth factor (IGF-1). The differentiated endothelial cells were exam...
